红松是濒危的珍稀植物,同时又是典型的外生菌根树种。论文对小兴安岭红松林内外生菌根真菌资源进行了调查,同时利用响应面法筛选优化厚环粘盖牛肝菌(Suillus grevillei)培养基的组分。经对小兴安岭红松林下外生菌根真菌采集调查与分类鉴定,初步确定有5个科、8个属2、1个种的真菌为红松外生菌根真菌类群,其中常见的有厚环粘盖牛肝菌、灰鹅膏菌(Amanita vaginata)、赭盖红菇(Russula mustelina)等。以改良MMN培养基为基础培养基,利用响应面法优化筛选确定S.grevillei的最佳培养基配方是KH2PO4 0.3 g、Peptone 16 g、Glucose 12 g、Beef ex-tract 15 g、CaCl2 0.05 g、MgSO4.7H2O 0.15 g、Malt extract 3 g、FeCl3(1%溶液)1.2 ml、NaCl0.025 g(、NH4)2HPO4 0.25 g、Vitamin B1微量(0.005 g)、Agar 20 g并加水至1 000 ml。该配方为S.grevillei菌剂的规模化生产奠定了基础。
Korean pine is an endangered rare plant, but also a typical ectomycorrhizal species. In this paper, the mycorrhizal fungi resources of Korean pine forest in Xiaoxing’anling were surveyed, and the components of Suillus grevillei medium were optimized by response surface methodology. After investigation and identification of ectomycorrhizal fungi collected from Pinus koraiensis forest in Xiaoxing’anling, five families and 8 genus and 2 species of fungi were identified as ectomycorrhizal fungi. Among them, there were common thick rings Capped bolete, Amanita vaginata, Russula mustelina and the like. The optimal culture medium of S.grevillei was identified by modified response surface methodology (MMN) as the basal medium. The best medium for S. gravillei was 0.3 g of KH2PO4, 16 g of Glucose, 12 g of Glucose, 15 g of Beef ex-tract, 0.05 g of CaCl2, 1.2 g of FeCl3 (1% solution), 0.25 g of NaCl0.025 g (, NH4) 2HPO4, 0.005 g of Vitamin B1, 20 g of Agar, and water to 1000 ml. The formula lays the foundation for the large-scale production of G. gravillei agents.
