目的:建立短暂外周血超抗原金黄色葡萄球菌A肠毒素(SEA)反应T细胞系,研究SEA诱导的T细胞凋亡。方法;采用超抗原SEA刺激PBMC增殖,rIL-2维持细胞生长,高速度Ficoll离心去除死细胞建立细胞系。FCM及琼脂糖凝胶电泳检测SEA诱导短期外周血SEA反应T细胞系凋亡。结果:SEA0.5~5μg/ml,rIL-2200U/ml能有效建立SEA反应T细胞系,此细胞系能在体外维持生长40d,对SEA的再次刺激有明显应答能力。1μg/mlSEA诱导同样剂量建立的SEA反应T细胞凋亡,16h能明显观察到凋亡特征。结论:SAg与rIL-2能成功建立短期SAg反应T细胞系,受到SEA再次作用后T细胞可发生凋亡。
Objective: To establish a transient peripheral blood superantigen Staphylococcus aureus enterotoxin (SEA) reactive T cell line to study the SEA induced T cell apoptosis. METHODS: The proliferation of PBMC was stimulated by superantigen SEA, rIL-2 cells were maintained for growth, and the dead cells were established by high speed Ficoll centrifugation. SEA-induced Apoptosis of SEA-reactive T Cell Line in Short-term Peripheral Blood Using FCM and Sepharose. Results SEA 0.5 ~ 5μg / ml and rIL-2200U / ml could effectively establish SEA-reactive T cell line. The cell line can maintain 40 days in vitro and has obvious response to SEA re-stimulation. 1μg / ml SEA induced the same dose of SEA-induced T cell apoptosis, 16h obvious apoptotic characteristics can be observed. Conclusion: SAg and rIL-2 can successfully establish short-term SAg-reactive T cell line, which can be T-cell apoptosis induced by SEA.
