目的研究新疆荒地阿魏Ferula syreitschikowii种质资源的亲缘关系及遗传多样性。方法以96份新疆荒地阿魏种质资源为材料,从33条ISSR引物中筛选出条带清晰、多态性好的引物15条,进行ISSR分子标记分析。结果共扩增获得292条完整、清晰的谱带,其中多态性条带为289条,多态位点比率为99.01%,表明新疆荒地阿魏种质资源多态性较高,具有较高的遗传多样性。经相关统计软件计算结果分析,平均有效等位基因数为1.725 6,平均Nei’s基因多样性指数为0.404 8,平均Shannon’s信息指数为0.587 9,遗传相似性系数为0.500 0~0.828 7。UPGMA聚类结果显示96份材料分为2个类群13个亚类,能够将不同来源的种质资源区分开来。结论从分子水平上揭示了新疆荒地阿魏地理分布与种质资源间的亲缘关系及较高的遗传多样性,为荒地阿魏品种鉴定提供相关依据。
Objective To study the genetic relationship and genetic diversity of Ferula syreitschikowii germplasm resources in the wasteland of Xinjiang. Methods 96 wild Xinjiang Uygur Autonomous Region germplasm resources were screened out from 33 ISSR primers. Fifteen primers with clear bands and good polymorphism were screened for ISSR molecular marker analysis. Results A total of 292 complete and clear bands were obtained by amplification, of which 289 were polymorphic bands and the percentage of polymorphic loci was 99.01%, which indicated that the genetic diversity of wild germplasm resources in Xinjiang was higher and higher Genetic diversity. The average number of effective alleles was 1.725 6, the average Nei’s gene diversity index was 0.404 8, the average Shannon’s information index was 0.587 9, and the genetic similarity coefficient was 0.500 0 ~ 0.828 7. UPGMA clustering results showed that 96 materials were divided into 13 subgroups of 2 groups, which could distinguish germplasm resources from different sources. Conclusion The phylogenetic relationship and genetic diversity of the wild geese in Xinjiang Uygur Autonomous Region were revealed at the molecular level, which provided the basis for the identification of the Asiatic wild Wei species.
