c-jun氨基末端激酶(JNK)是丝裂原活化蛋白激酶(MAPK)家族的成员。JNK信号转导通路参与细胞的胚胎发育、分化、凋亡等。JNK相互作用蛋白-1/(JIP-1)/IB1(islet-brain-1)是哺乳动物JNK信号转导通路的支架蛋白,通过蛋白质之间的相互作用对JNK信号转导通路起关键的调节作用。JNK信号转导通路参与β细胞凋亡、胰岛素基因表达障碍和胰岛素抵抗的发生机制。通过改变细胞中JIP-1的含量等方法抑制JNK的活性或干扰JNK与胰岛素受体底物(IRS)-1的相互作用可成为糖尿病治疗的新靶点。
The c-jun amino-terminal kinase (JNK) is a member of the mitogen-activated protein kinase (MAPK) family. JNK signal transduction pathway involved in embryonic development, differentiation, apoptosis and so on. JNK-interacting protein-1 (JIP-1) / IB1 (islet-brain-1) is a scaffold protein of mammalian JNK signaling pathway and plays a key role in regulating the JNK signal transduction pathway through protein-protein interactions effect. JNK signal transduction pathway is involved in the pathogenesis of β-cell apoptosis, impaired insulin gene expression and insulin resistance. Inhibiting the activity of JNK or interfering with the interaction between JNK and insulin receptor substrate (IRS) -1 can be a new target for the treatment of diabetes by changing the content of JIP-1 in cells.
