目的考察延胡索在大鼠血浆中的药动学特征及炮制对其吸收和代谢的影响。方法采用高效液相测定灌胃延胡索提取物后的大鼠血浆中的药物浓度。结果建立了同时测定大鼠血浆中延胡索乙素,原阿片碱和去氢紫堇碱的方法,该方法的低,中,高3种药物浓度的回收率,日内和日间精密度均符合要求。结论醋炙能加快延胡索乙素和去氢紫堇碱在体内的吸收,同时延缓二者的消除,其中对延胡索乙素的作用更明显,而对原阿片碱影响不大;酒炙对延胡索乙素的作用与醋炙相似,但作用较醋炙弱,同时对原阿片碱和去氢紫堇碱均有延缓消除的作用。
Objective To investigate the pharmacokinetics of Corydalis yanhusuo in rat plasma and the effects of concocting on its absorption and metabolism. Methods The concentration of drug in rat plasma after intragastric administration of extract of Corydalis yanhusuo was determined by HPLC. Results A method for the simultaneous determination of tetrahydropalmatine, protopine and dehydrovioside was established in rat plasma. The recoveries of the three concentrations of low, medium, and high concentrations of the drug were achieved. The intra-day and inter-day precisions met the requirements. . Conclusion Acetazolidine can accelerate the absorption of tetrahydropalmatine and dehydroviopuricine in vivo, and delay the elimination of the two. The effect of tetrahydropalmatine on tetrahydropalmatine is more obvious, and the effect of acetamidine on protopine is not significant. The effect of quinone is similar to that of vinegar, but its effect is weaker than that of vinegar. At the same time, it can delay the elimination of the original opiate and dehydroviodamine.
