目的 :研究“益肝浓缩煎剂”对四氯化碳大鼠肝纤维化预防与治疗作用 ,并对其机制进行初步探讨。方法 :以CCI4 诱导大鼠肝纤维化模型 ,造模前2周及结束后分别予“益肝浓缩煎剂”进行防治 ,各持续10周 ;测定肝组织羟脯氨酸 (Hyp)、丙二醛(MDA)含量 ,光镜、电镜观察肝脏病理形态变化。结果 :“益肝浓缩煎剂”预防、治疗组Hyp、MDA显著降低 (P<0 01) ,胶原沉积及脂肪变性明显减轻 (P<0 05) ;此外 ,“益肝浓缩煎剂”预防组α -平滑肌肌动蛋白表达减少。结论 :“益肝浓缩煎剂”对大鼠肝纤维化具有明确防治作用 ;其疗效作用机制可能与抗脂质过氧化和抑制肝星状细胞活化有关。
Objective : To study the preventive and therapeutic effects of “Yigan decoction concentrate” on liver fibrosis in rats with carbon tetrachloride, and to discuss its mechanism. METHODS: Rat models of liver fibrosis were induced by CCI4. Two weeks before and after model establishment, “Yigan decoction concentrate” was used for prevention and treatment respectively for 10 weeks. Liver tissue hydroxyproline (Hyp) and C2 were measured. Aldehyde (MDA) content, light microscopy and electron microscopy were used to observe the pathological changes of the liver. Results: Hyp and MDA in the prevention and treatment group of “Yigan Concoction decoction” decreased significantly (P <0 01), collagen deposition and steatosis were significantly reduced (P <0 05); In addition, “Yigan decoction concentrate” prevention group Alpha-smooth muscle actin expression decreased. Conclusion: “Yigan Concentrate Decoction” has a definite preventive and therapeutic effect on liver fibrosis in rats; its therapeutic mechanism may be related to anti-lipid peroxidation and inhibition of hepatic stellate cell activation.
