目的:探讨叶酸补充拮抗5,10-亚甲基四氢叶酸还原酶(MTHFR)基因下调的作用机制。方法:MTT法检测叶酸梯度补充后拮抗MTHFR基因功能下调对细胞增殖的影响;流式细胞仪分析MTHFR基因功能下调补充叶酸前后细胞周期的改变。结果:MTHFR基因功能下调后,不同剂量叶酸补充可以逆转MTHFR基因功能下调对EPM细胞的不良影响,影响程度与补充叶酸的浓度在一定范围内具有剂量依赖性。结论:MTHFR基因是重要的先天性唇腭裂候选基因,叶酸补充可以拮抗MTHFR基因功能下调对胚胎腭突发育的不良影响。
Objective: To explore the mechanism of folic acid supplementation antagonizing the down-regulation of 5,10-methylenetetrahydrofolate reductase (MTHFR) gene. Methods: MTT assay folic acid gradient supplementation antagonized the MTHFR gene function down-regulation on cell proliferation; MTHFR gene function by flow cytometry analysis of cell cycle changes before and after folic acid supplementation. RESULTS: After MTHFR gene function was down-regulated, different doses of folic acid supplementation could reverse the adverse effect of MTHFR gene function down-regulation on EPM cells. The degree of effect and the concentration of folic acid supplementation were dose-dependent in a certain range. CONCLUSION: MTHFR gene is an important congenital cleft lip and palate candidate gene. Folic acid supplementation can antagonize the adverse effects of MTHFR gene function down-regulation on embryonic palatal process.
