目的:研究白藜芦醇抑制辐射诱导的NLRP3表达的机理。方法:将间充质干细胞分为空白对照组、单纯照射组、白藜芦醇照射组、NAM照射组,应用Western blot和RT-PCR等方法检测IL-1β和NLRP3的转录和表达。结果:单纯照射组IL-1β和NLRP3转录与表达显著升高(t=20.96,P<0.05),白藜芦醇组与NAM组IL-1β和NLRP3转录与表达较照射组明显受到抑制(t=16.57,8.447;P<0.05)。结论:白藜芦醇能够明显抑制辐射诱导的IL-1β和NLRP3转录与表达,并且可能是通过SIRT1-NLRP3途径发挥作用的。
AIM: To investigate the mechanism of resveratrol in inhibiting radiation-induced NLRP3 expression. Methods: The mesenchymal stem cells were divided into blank control group, radiation group, resveratrol group and NAM irradiation group. The transcription and expression of IL-1β and NLRP3 were detected by Western blot and RT-PCR. Results: The transcription and expression of IL-1β and NLRP3 were significantly increased (t = 20.96, P <0.05) in the group of irradiation alone. The transcription and expression of IL-1β and NLRP3 in resveratrol group and NAM group were significantly inhibited = 16.57, 8.447; P <0.05). CONCLUSION: Resveratrol can significantly inhibit the transcription and expression of radiation-induced IL-1β and NLRP3, and may play a role through the SIRT1-NLRP3 pathway.
