目的:建立HPLC法测定肾通颗粒中淫羊藿配伍知母、生地后各配伍煎液的淫羊藿苷含量的分析方法,研究药材不同配伍对淫羊藿苷溶出量的影响。方法:采用AlltechC18柱(4.6mm×250mm,5μm),乙腈-0.1%冰醋酸(30∶70)为流动相,紫外检测波长270nm,柱温30℃,流速1.0mL·min-1。结果:该法简便、准确、灵敏,其他成分对测定无干扰,可用于配伍研究中淫羊藿苷的含量测定。结论:淫羊藿与知母、生地配伍后淫羊藿苷的溶出量有显著性下降;三者不同配伍比例对淫羊藿苷溶出量无显著性影响。
Objective: To establish an analytical method for the determination of icariin in Shentong Granules by HPLC and determine the content of icariin in each decoction after its combination with Anemarrhenae radix and Radix Astragali, and to study the effects of different combinations on the amount of icariin released from herbs. Methods: Alltech C18 column (4.6mm×250mm, 5μm), acetonitrile-0.1% glacial acetic acid (30∶70) as mobile phase, UV detection wavelength at 270nm, column temperature at 30°C, flow rate at 1.0mL·min-1. Results: The method is simple, accurate and sensitive. The other components have no interference with the determination. It can be used for the determination of icariin in compatibility studies. Conclusion: The dissolution of icariin was significantly decreased after compatibility between herba and anemarrhena as well as its habitat. There was no significant effect on the amount of icariin eluted from the three combinations.
