目的观察扶肾降浊法中药对系膜增生性肾炎大鼠肾小管间质成纤维细胞Ⅰ、Ⅲ型胶原(COLⅠ及COLⅢ)mRNA和蛋白的调节作用。方法采用扶肾降浊法中药水溶液灌胃Wistar大鼠常规制备含药血清,体外培养12、16、20周龄系膜增生性肾炎大鼠模型的肾间质成纤维细胞,采用实时荧光定量PCR法和Western blotting法研究扶肾降浊法含药血清对病理状态间质成纤维细胞COLⅠ及COLⅢmRNA和蛋白表达的影响。结果系膜增生性肾炎模型大鼠肾间质成纤维细胞的COLⅠ及COLⅢmRNA和蛋白表达上调(P<0.05),扶肾降浊法含药血清能下调病理状态肾间质成纤维细胞的COLⅠ及COLⅢmRNA和蛋白表达(P<0.05)。结论扶肾降浊法含药血清可下调病理状态肾间质成纤维细胞的COLⅠ及COLⅢmRNA表达,减少COLⅠ及COLⅢ蛋白表达,对肾间质成纤维细胞具有保护作用。
Objective To observe the regulatory effect of traditional Chinese medicine Fufang Jiangzhuo on mRNA and protein expressions of type Ⅰ and type Ⅲ collagen (COL Ⅰ and COL Ⅲ) in mesangial proliferative glomerulonephritis rats. Methods Wistar rats were treated with Fuzheng Jiangzhuo aqueous solution of traditional Chinese medicine and routinely prepared drug-containing serum. Rat renal interstitial fibroblasts of 12, 16 and 20 weeks old rats were cultured with real-time fluorescence quantitative PCR The effects of Fufang Jiangzhuo containing serum on the expression of COL Ⅰ and COL Ⅲ mRNA and protein in the pathological state of interstitial fibroblasts were studied by Western blotting and Western blotting. Results The mRNA and protein expressions of COL Ⅰ and COL Ⅲ were increased in mesangial fibroblasts of mesangial proliferative glomerulonephritis model rats (P <0.05). The serum containing Fuzhu Jiangzhuo Decoction could decrease the COLⅠ COL Ⅲ mRNA and protein expression (P <0.05). Conclusion Fufang Jiangzhuo containing serum can reduce the expression of COL Ⅰ and COL Ⅲ mRNA in the pathological state of renal interstitial fibroblasts, reduce the expression of COL Ⅰ and COL Ⅲ protein, and have a protective effect on renal interstitial fibroblasts.
