为探讨白细胞介素 -6(IL -6)对人脐静脉内皮细胞 (HUVEC)黏附分子表达的影响。我们采用培养的原代HUVEC为实验对象 ,将其分为 3组 ,分别为空白对照组、小剂量IL -6组 ( 50U ml)、大剂量IL-6组 ( 1 0 0U ml) ,应用免疫细胞化学染色技术检测细胞间黏附分子 1 (ICAM -1 )及血管细胞黏附分子 1(VCAM -1 )的表达。结果 :IL -6组内皮细胞ICAM -1及VCAM -1的表达较对照组明显增加。对照组阳性率分别为 7 0 %± 1 2 %、1 5%± 0 5% ,小剂量IL -6组分别为 2 0 5%± 2 3 %、1 9 7%± 1 9% ,大剂量IL -6组为 2 6 3 %± 3 1 %、2 5 0 %± 2 3 % ( P <0 0 5)。作者认为 ,IL -6能够促进内皮细胞ICAM -1及VCAM -1的表达并呈剂量依赖关系。
To investigate the effect of interleukin-6 (IL-6) on the expression of adhesion molecules in human umbilical vein endothelial cells (HUVECs). We used cultured primary HUVECs as experimental subjects and divided them into three groups: blank control group, low dose IL-6 group (50U ml) and high dose IL-6 group (100U ml) Cytochemical staining was used to detect the expression of intercellular adhesion molecule 1 (ICAM -1) and vascular cell adhesion molecule 1 (VCAM -1). Results: The expressions of ICAM - 1 and VCAM - 1 in endothelial cells of IL - 6 group were significantly higher than those in control group. The positive rates of the control group were 70% ± 1 2% and 15% ± 0 5%, respectively, while those of the low dose IL-6 group were respectively 20 5% ± 23% and 1 9 7% ± 1 9% The IL-6 group was 26.3% ± 31.1%, 25.0% ± 23% (P <0.05). The authors believe that IL-6 can promote the expression of ICAM-1 and VCAM-1 in a dose-dependent manner.
