目的:探讨尿激酶型纤溶酶原激活物(uPA)在各种大肠癌细胞的表达及其意义。方法:采用Real time PCR检测大肠癌细胞HT29,HCT116,SW1116,SW480等细胞系中uPA的mRNA表达,Western blot检测uPA的蛋白表达水平并进行比较分析。结果:在转录水平,SW480细胞中uPA mRNA表达最高,其次是HT29细胞,另外两种细胞表达相对低。uPA的蛋白表达的情况表明,SW480细胞中uPA的表达较其他组明显高(p<0.05),而SW1116细胞中几乎未见uPA的蛋白表达。结论:SW480细胞中uPA的表达较其他细胞系高,这可能与肿瘤细胞转移能力强弱有关。
Objective: To investigate the expression of urokinase-type plasminogen activator (uPA) in various colorectal cancer cells and its significance. Methods: Real-time PCR was used to detect the mRNA expression of uPA in colorectal cancer cell lines such as HT29, HCT116, SW1116 and SW480. The protein expression of uPA was detected by Western blot and compared. Results: At the transcriptional level, uPA mRNA was highest in SW480 cells, followed by HT29 cells, and the other two cells were relatively low. The expression of uPA protein showed that the expression of uPA in SW480 cells was significantly higher than that of other groups (p <0.05), but almost no expression of uPA in SW1116 cells. Conclusion: The expression of uPA in SW480 cells is higher than other cell lines, which may be related to the metastasis ability of tumor cells.
