[目的]探讨三磷酸腺苷-氯化镁(ATP-MgCl_2)对新生大鼠高氧肺纤维化的影响.[方法]选用同孕母大鼠正常出生后第3d的新生Wistar大鼠,随机分为正常环境对照组、氧体积分数高模型组及ATP-MgCl_2治疗组.正常环境对照组暴露于室内空气中,其余2组暴露于氧体积分数大于85%的环境中,共14d.于出生后第2d起每日腹腔注射给予ATP-MgCl_2治疗组ATP-MgCl_2 45mg/kg,其余2组均腹腔注射给予等剂量生理盐水.于指定天数时每组随机抽出10只新生鼠,利用生理盐水及10%(质量分数)甲醛行心脏灌流后取整个肺,固定,制片,在光学显微镜下观察肺泡及间质等相关组织的病理改变;利用免疫组织化学染色法观察肺组织中干扰素-γ(IFN-γ)及基质金属蛋白酶-9(MMP-9)的表达水平.[结果]正常环境对照组IFN-γ未见显色,而第7 d时氧体积分数高模型组IFN-γ阳性表达明显增高,第14 d时达到高峰,ATP-MgCl_2治疗组IFN-γ表达强度在各时间段均明显低于氧体积分数高模型组(P<0.01).正常环境对照组MMP-9仅在肺组织中散在表达,而第7 d时氧体积分数高模型组MMP-9显色表达强度增加,第14 d时表达最强,ATP-MgCl_2治疗组MMP-9表达强度在各时间段均明显低于氧体积分数高模型组(P<0.01).[结论]ATP-MgCl_2对新生大鼠高氧肺组织纤维化具有保护作用.
[Objective] To investigate the effect of adenosine triphosphate-magnesium chloride (ATP-MgCl_2) on hyperoxic pulmonary fibrosis in neonatal rats. [Methods] Newborn Wistar rats were randomly divided into normal control Group, high oxygen concentration model group and ATP-MgCl 2 treatment group.The normal environment control group was exposed to the indoor air, the other two groups were exposed to the environment of the oxygen volume fraction greater than 85% for a total of 14 days.On the second day after birth ATP-MgCl 2 45mg / kg was given intraperitoneally to the rats in the ATP-MgCl 2 treatment group, and the other two groups were injected intraperitoneally with the same dose of normal saline.On the given days, 10 newborn rats were randomly selected in each group and treated with normal saline and 10% ) Formaldehyde was collected from the heart after perfusion of the heart, fixed, and made into pieces. The pathological changes of alveolar and interstitial tissues were observed under a light microscope. The expression of IFN-γ in the lung tissue was observed by immunohistochemical staining. And MMP-9 (MMP-9). [Results] No expression of IFN-γ was observed in the normal control group, but IFN-γ expression in model group with high oxygen concentration was significantly increased on the 7th day. Peaked on day 14, and IFN-γ expression in ATP-MgCl 2 treatment group The intensity of MMP-9 was significantly lower in model group than that in model group (P <0.01) at all time points.The expression of MMP-9 in normal control group was only sporadically expressed in lung tissue, while on the 7th day, The expression of MMP-9 in ATP-MgCl 2 group was significantly lower than that in high-oxygen group at each time point (P <0.01). [Conclusion] ATP-MgCl 2 On neonatal rats with high oxygen lung tissue fibrosis has a protective effect.
