观察肌肉接种编码恶性疟原虫复合多价保护性抗原的重组质粒pCDNA3—Pfs诱导小鼠产生的免疫血清对体外培养恶性疟原虫的抑制作用。方法将pCDNA3—Pfs通过肌肉途径免疫接种BALB/c小鼠,3次免疫后制备抗血清.进行酶联免疫吸附试验和疟原虫体外生长抑制试验。结果血清特异抗体滴度达1:2560,体外抑制试验示随培养物中免疫血清浓度的增加及孵育时间的延长而抑制程度增强,l:5稀释作用于原虫72h后,抑制率可达68.7%。结论直接肌肉接种质粒pCDNA3—Pj’8可诱导BALB/C小鼠产生良好的免疫反应及明显的抑制原虫体外生长发育的作用。
To observe the inhibitory effect of intramuscular immunization of sera induced by recombinant plasmid pCDNA3-Pfs encoding P. falciparum multivalent protective antigen on P. falciparum in vitro. Methods BALB / c mice were immunized with pCDNA3-Pfs via the muscle route and antisera were prepared after three immunizations. Enzyme-linked immunosorbent assay and Plasmodium in vitro growth inhibition test. Results The titer of serum specific antibody reached 1: 2560. The in vitro inhibition test showed that the inhibition was enhanced with the increase of serum concentration and incubation time in culture, and the inhibitory rate was 68 after 1: 5 dilution for 72h. 7%. Conclusion Direct intramuscular inoculation of plasmid pCDNA3-Pj’8 can induce good immune response in BALB / C mice and significantly inhibit the growth and development of protozoa in vitro.
