目的 :观察丹皮酚 (Pae)对人胎儿主动脉平滑肌细胞增殖的抑制作用。方法 :利用体外细胞培养技术 ,用人高脂血清造成平滑肌细胞 (SMC)增殖的模型并通过 MTT测定、细胞计数及透射电镜下的细胞超微结构的观察等方法 ,观察了 Pae对人胎儿平滑肌细胞的保护作用。结果 :人高脂血清可明显刺激人胎儿平滑肌细胞的增殖并引起其表型的改变 ,而 Pae可呈浓度依赖性地抑制平滑肌细胞的增殖并可显著抑制高脂血清对 SMC的促增殖作用。结论 :Pae抗动脉粥样硬化的作用可能部分是由于抑制 SMC异常增殖的结果。
Objective: To observe the inhibitory effect of Pae on the proliferation of human fetal aorta smooth muscle cells. METHODS: Using in vitro cell culture technique, human hyperlipidemia serum was used to induce the proliferation of smooth muscle cells (SMC) and the effects of Pae on human fetal smooth muscle cells were observed by MTT assay, cell counting, and ultrastructure observation under transmission electron microscope. Protection. RESULTS: Human hyperlipidemia significantly stimulated the proliferation of human fetal smooth muscle cells and caused its phenotypic changes, while Pae inhibited the proliferation of smooth muscle cells in a concentration-dependent manner and significantly inhibited the pro-proliferative effects of hyperlipidemia on SMC. Conclusion : The effect of Pae antiatherosclerosis may be partly due to the inhibition of abnormal proliferation of SMC.
