目的:探讨血管紧张素Ⅱ受体阻断剂在体外对肾小球系膜细胞及I型胶原基因启动子的作用。方法:大鼠肾小球系膜细胞株传代培养。BrdU掺入系膜细胞,加入各浓度los24小时后,经固定、变性及抗BrdU酶标抗体作用,ELISA法测定洛沙坦对细胞增殖的影响。FuGENE转染试剂转染系膜细胞,CAT-ELISA测定洛沙坦对两种重组体的影响。结果:在10%血清浓度1640培养基条件下,洛沙坦作用浓度从10-7-10-5M作用24小时后各组细胞增殖差异显著。洛沙坦作用于两种重组体24小时后,CAT测定结果提示:两种重组体转染细胞后,洛沙坦组与对照组的CAT值,差异也有显著性。结论:血管紧张素Ⅱ受体阻断剂洛沙坦对肾小球系膜细胞增殖有明显的抑制作用,并对I型胶原基因启动子具有负性调控作用。
Objective: To investigate the effect of angiotensin Ⅱ receptor blockers on mesangial cells and type I collagen gene promoter in vitro. Methods: Rat mesangial cell lines were subcultured. BrdU was incorporated into mesangial cells. After adding losartan at various concentrations for 24 hours, the effect of losartan on cell proliferation was determined by ELISA. The mesangial cells were transfected with FuGENE transfection reagent. The effects of losartan on the two recombinants were determined by CAT-ELISA. Results: Under the conditions of 10% serum concentration of 1640 medium, the proliferation of cells in each group was significantly different after the concentration of losartan was increased from 10-7-10-5M for 24 hours. Losartan effect on two recombinants after 24 hours, CAT measurement results suggest that: two recombinant transfected cells, losartan group and the control group of CAT values, the difference was also significant. CONCLUSION: Losartan, an angiotensin II receptor blocker, significantly inhibits the proliferation of glomerular mesangial cells and negatively regulates type I collagen gene promoter.
