目的 探讨年龄对光化学诱导的局灶性脑缺血皮层神经元线粒体形态结构的影响。方法 对老年和青年大鼠缺血后 4小时、2 4小时、5天缺血边缘区行电镜观察 ,通过计算机图象分析系统对神经元线粒体进行形态计量分析。结果 老年组神经元线粒体数密度和比表面较青年组小 (P <0 .0 5~ 0 .0 1)、而平均体积增大。在相同时间点的缺血边缘区 ,老年组神经元线粒体平均体积、圆球度均较青年组大 ,而比表面减小。两组线粒体平均体积、圆球度及体密度在缺血 4小时增加、2 4小时更明显 ,5天后减小 ,比表面的变化则相反(P <0 .0 5 )。结论 由于老年大鼠存在线粒体老化等年龄相关性改变 ,在同等缺血打击下 ,线粒体肿胀较青年大鼠更严重 ,修复能力更低
Objective To investigate the effect of age on the mitochondrial morphology and structure of cortical neurons induced by photochemistry in focal cerebral ischemia. Methods Electron microscopy was performed on ischemic border regions 4 hours, 24 hours and 5 days after ischemia in aged and young rats. Morphometric analysis of mitochondria in neurons was performed by computer image analysis system. Results The number density and specific surface area of mitochondria in the neurons in the aged group were smaller than those in the young group (P <0.05-0.01), while the average volume was increased. In the ischemic marginal zone at the same time point, the average mitochondrial volume and sphericity of the neurons in the aged group were larger than those in the young group, and the specific surface area was decreased. The average volume, sphericity and body density of mitochondria in both groups increased at 4 hours after ischemia, more obviously at 24 hours, and decreased at 5 days, and the changes of specific surface area were opposite (P <0.05). Conclusions Due to age-related changes such as mitochondrial aging in aged rats, mitochondrial swelling is more severe and less likely to be repaired in young rats under the same ischemic attack
