目的:研究安息香提取物对损伤内皮细胞中炎性介质乳酸脱氢酶(LDH)、肿瘤坏死因子(TNF-α)及白细胞介素-8(IL-8)活性的调节作用,探究安息香治疗动脉粥样硬化的作用机制。方法:应用补体蛋白纯品于体外在人脐静脉内皮细胞表面组装亚溶破剂量的C5b-9,建立内皮损伤模型,设立安息香提取物高、中、低剂量组,苯甲酸高、中、低剂量组,模型对照组及空白对照组,分别用不同浓度的安息香提取物及苯甲酸标准品处理细胞,应用紫外分光光度法及Elisa试剂盒测定细胞培养上清液中LDH、TNF-α及IL-8浓度。结果:安息香提取物高、中、低剂量组与模型对照组比较,LDH及IL-8分泌水平差异均有统计学意义(P<0.01);安息香提取物高剂量组中LDH及IL-8的分泌水平与空白对照组水平接近,差异无统计学意义(P>0.05);与模型对照组比较,安息香提取物中剂量组TNF-α分泌水平显著降低(P<0.01)。结论:安息香可能通过降低炎性介质LDH、TNF-α及IL-8活性,从而修复内皮细胞损伤。
Objective: To study the effects of benzoin extract on the activity of inflammatory mediators of lactate dehydrogenase (LDH), tumor necrosis factor (TNF-α) and interleukin-8 (IL-8) Atherosclerosis mechanism of action. Methods: The sub-soluble dosage of C5b-9 was assembled on the surface of human umbilical vein endothelial cells by using the pure product of complement protein. The model of endothelial injury was established. The high, medium and low dose benzoic acid extract groups were established. Dose group, model control group and blank control group were treated with different concentrations of benzoin extract and benzoic acid standard cells, UV spectrophotometry and Elisa kit determination of cell culture supernatant LDH, TNF-α and IL -8 concentration. Results: Compared with the model control group, the levels of LDH and IL-8 in the benzoin extract of high, medium and low dose groups were significantly different (P <0.01); the levels of LDH and IL-8 Compared with the control group, the secretion of TNF-α in middle dose benzoin group was significantly lower (P <0.01). Conclusion: Benoban may repair endothelial cell injury by decreasing the activity of inflammatory mediators LDH, TNF-α and IL-8.
