目的分析引起吉林省2011年手足口病(Hand,foot and mouth disease,HFMD)流行的肠道病毒71型(Enterovirus 71,EV71)VP1区基因的特征。方法对2011年分离自吉林省8个市(州)HFMD病例的35株EV71分离株的VP1编码区基因进行RT-PCR扩增及序列测定,应用Bioedit和MEGA 4.0软件对VP1基因进行同源性和基因亲缘关系分析。结果吉林省2011年的35株EV71分离株均属于C4a基因亚型;其VP1区基因的核苷酸序列同源性在95.9%~100%之间,分属于3个进化枝,形成3个传播链,与2008年安徽阜阳株亲缘关系最近,VP1区基因核苷酸序列同源性达97.6%~99.3%,与2007年的北京株和2003年的浙江株亲缘关系相对较远;2011年吉林省各市中,既存在3个EV71传播链共流行的市,同时监测到2个EV71传播链共流行的市,还发现3个只有单一EV71传播链的市,每个传播链均在5个市流行传播。结论 2011年在吉林省内,3个C4a基因亚型EV71的传播链同时循环传播引起HFMD的流行,未发现优势传播链。
Objective To analyze the characteristics of the VP1 gene of Enterovirus 71 (EV71) that caused the epidemic of Hand, Foot and Mouth Disease (HFMD) in Jilin Province in 2011. Methods The VP1 coding region of 35 strains of EV71 isolates from 8 cities (prefectures) in Jilin province in 2011 were amplified by RT-PCR and sequenced. The homology of VP1 gene was analyzed by Bioedit and MEGA 4.0 software And genetic relationship analysis. Results All the 35 EV71 isolates in Jilin Province in 2011 belonged to C4a subtype. The nucleotide sequence homology of the VP1 gene was between 95.9% and 100%, belonging to 3 clades, forming 3 spreads Recently, the nucleotide sequence homology of VP1 gene was 97.6% -99.3%, which was relatively distant from Beijing strain in 2007 and Zhejiang strain in2007. Jilin strain Among the cities and provinces in the province, there are three cities with common epidemic of EV71 transmission chain and two cities with epidemic prevalence of EV71 transmission line at the same time. Three cities with only single EV71 transmission chain were also found, each with 5 transmission lines Pop spread. Conclusions In Jilin Province in 2011, the circulation of the three C4a subtypes of EV71 circulating at the same time caused the epidemic of HFMD. No dominant transmission chain was found.
