目的 了解羟基喜树碱 (HCPT )对内皮细胞生长的影响 ,进而了解其对血管生成的抑制作用。方法 ECV 3 0 4细胞体外培养并检测不同浓度HCPT对细胞增殖率、形态、粘附功能的影响 ,采用流式细胞仪测定细胞凋亡指数和细胞周期。结果 终浓度为 4.0、7.5、15 .0、3 0 .0、60 .0ng/ml的HCPT对ECV 3 0 4的增殖抑制率分别为 6.7%、3 7.9%、69.5 %、87.4%、91.0 % ;7.5ng/ml和 15 .0ng/ml浓度处理的细胞凋亡率分别为 2 0 .0 %和 3 5 .1% ;细胞的形态和贴壁能力均有明显改变。 结论 HCPT可抑制内皮细胞增殖 ,并能使其形态改变 ,粘附功能下降 ,显示了其抗血管生成的潜能。
Objective To investigate the effect of HCPT on the growth of endothelial cells and to investigate its inhibitory effect on angiogenesis. Methods ECV 3 0 4 cells were cultured in vitro and the effects of different concentrations of HCPT on cell proliferation rate, morphology and adhesion function were detected. The apoptosis index and cell cycle were determined by flow cytometry. Results The inhibitory rates of HCPT at the concentrations of 4.0, 7.5, 15.0, 3.0, 0.0, 60.0ng / ml on ECV304 were 6.7%, 39.9%, 69.5%, 87.4%, 91.0% The apoptotic rates of 7.5ng / ml and 15.0ng / ml were 20.0% and 35.1%, respectively. The morphological and adherent ability of cells were significantly changed. Conclusion HCPT can inhibit the proliferation of endothelial cells, change the morphology and decrease the adhesion function of HCPT, indicating its anti-angiogenic potential.
