为探讨特异性抗体解除洋地黄毒性作用的机制,应用由淋巴细胞杂交瘤技术制备的高亲和力抗地高辛特异性MAb,研究了特异性抗体对地高辛抑制家兔红细胞(RRBC)膜Na~+,K~+-ATP酶活性的拮抗作用。实验结果表明,特异性MAb FD8可高效拮抗地高辛引起的Na~+,K~+-ATP酶活性抑制。10~(4·26)稀释的FD8 MAb腹水在试管内可使4ng/ml地高辛引起的酶活性抑制恢复达50%。给地高辛中毒动物静脉注射FD8 MAb腹水后,RBC膜Na~+,K~+-ATP酶活性抑制迅速恢复,RBC内Na~+、K~+离子浓度异常也得到恢复。本研究从受体水平证实了特异性MAb解除地高辛毒性的作用机制。
In order to explore the mechanism of specific antibodies to relieve the toxicity of digitalis, the high affinity anti-digoxin-specific MAb prepared by lymphocyte hybridoma technique was used to study the inhibitory effect of digoxin on rabbits red blood cells (RRBC) membrane Na ~ +, K ~ + -ATPase activity antagonism. Experimental results show that specific MAb FD8 can effectively inhibit digoxin-induced inhibition of Na ~ +, K ~ + -ATPase activity. FDG MAb ascites diluted 10 ~ (4.26) in the test tube inhibited digoxin-induced inhibition of 4ng / ml recovery by up to 50%. After intravenous injection of FD8 MAb ascites into digoxin-poisoned animals, the inhibition of Na +, K + -ATPase activity of RBC membrane restored rapidly, and the Na +, K + ion concentration abnormalities in RBC recovered. In this study, the mechanism of action of specific MAbs to relieve digoxin toxicity was demonstrated at the receptor level.
