为研究超声波对啤酒废酵母中超氧化物歧化酶(SOD)的辅助提取效果,以总酶活和比酶活作为指标研究液料比、超声功率、超声间歇时间、超声总时间对破壁效果的影响。在单因素试验的基础上进行3因素3水平的BoxBehnken试验设计,回归模型预测的最优参数为液料比3.7︰1(mL/g)、间歇时间5 min、超声总时间4.5 min,验证值7 551.7 U和预测值7 550.9 U非常接近,回归系数R2=0.976 7,说明试验得到的回归模型能较好的应用于超声辅助提取酵母中SOD工艺参数和总酶活的预测。
In order to study the effect of ultrasound on the extraction of superoxide dismutase (SOD) in beer waste yeast, the effects of liquid material ratio, ultrasonic power, ultrasonic intermittent time and total ultrasonic time on the effect of broken wall were studied by using total enzyme activity and specific enzyme activity influences. The BoxBehnken experimental design with 3 factors and 3 levels based on the single factor experiment was carried out. The optimal parameters of the regression model were the liquid to solid ratio of 3.7︰1 (mL / g), intermittent time of 5 min, total ultrasonic time of 4.5 min, 7 551.7 U and the predicted value of 7 550.9 U are very close, the regression coefficient R2 = 0.976 7, indicating that the regression model obtained by the experiment can be better applied to the prediction of the SOD process parameters and the total enzyme activity in the ultrasonic assisted extraction of yeast.
