目的:建立高效液相色谱(HPLC)同时测定广藿香中雷杜辛黄酮醇和广藿香酮的方法,对11批市售的广藿香药材进行质量分析。方法:采用高效液相色谱法。Phenomenex Luna C18色谱柱(4.6 mm×250 mm,5μm),流动相乙腈-0.2%甲酸溶液(65∶35),等度洗脱;检测波长310 nm,流速0.8 mL.min-1,柱温30℃。结果:雷杜辛黄酮醇在0.020~0.36μg,广藿香酮在0.025~0.40μg呈良好线性关系,相关系数均为1.000 0,平均回收率雷杜辛黄酮醇为98.57%(RSD 2.06%),广藿香酮为100.04%(RSD 2.66%);11批样品中雷杜辛黄酮醇含量在1.13~8.72 mg.g-1,广藿香酮含量在1.19~4.38 mg.g-1。结论:方法简便、快速、准确,重复性好,可用于广藿香中雷杜辛黄酮醇及广藿香酮含量的同时测定。
Objective: To establish a HPLC method for the simultaneous determination of patchouli flavonol and patchoulenone in patchouli, and to analyze the quality of patchouli medicinal materials in 11 batches. Methods: Using high performance liquid chromatography. Phenomenex Luna C18 column (4.6 mm × 250 mm, 5 μm), mobile phase of acetonitrile-0.2% formic acid solution (65:35), isocratic elution was performed at a detection wavelength of 310 nm with a flow rate of 0.8 mL · min- ℃. Results: The results showed that there was a good linear relationship between 0.020-0.36 μg of flavodione and 0.025-0.40 μg of patchoulenone with the correlation coefficients of 1.000 0. The average recoveries of flavodoxin were 98.57% (RSD 2.06%). , And patchouli ketone was 100.04% (RSD 2.66%). The contents of flavodione in the 11 batches of samples were 1.13 ~ 8.72 mg.g-1 and patchouli ketone contents were 1.19 ~ 4.38 mg.g-1. Conclusion: The method is simple, rapid, accurate and reproducible. It can be used for the simultaneous determination of flavonol and patchoulenone in patchouli.
