目的:研究钙调神经磷酸酶(CaN)对血管钙化的调节作用。方法:4周龄SD雄性大鼠36只,随机分为对照组、钙化组和环孢霉素A(cyclosporine A,CsA)组,采用华法林和维生素D3皮下注射方法建立主动脉钙化模型,免疫组织化学方法测定主动脉CaN蛋白表达,原位杂交方法测定主动脉CaNAα mRNA的表达。结果:与对照组比较,钙化组大鼠主动脉CaN蛋白、CaNAαmRNA表达明显增加(P<0.01,P<0.01),在CsA组则表达减少(P<0.05,P<0.01)。结论:CaN可能通过Aα对动脉钙化进行调节。
Objective: To investigate the regulation of calcineurin (CaN) on vascular calcification. Methods: Thirty-six SD male rats aged 4 weeks were randomly divided into control group, calcification group and cyclosporine A (CsA) group. Aortic calcification models were established by subcutaneous injection of warfarin and vitamin D3, Aortic CaN protein expression was detected by immunohistochemistry and expression of CaNαα mRNA in the aorta was detected by in situ hybridization. Results: Compared with the control group, the expressions of CaN protein and CaNαmRNA in the aorta of rats in calcification group were significantly increased (P <0.01, P <0.01), and decreased in CsA group (P <0.05, P <0.01). Conclusion: CaN may regulate arterial calcification through Aα.
