研究了二乙基亚硝胺诱发大鼠肝癌过程中1,2-甘油二脂(DAG)、磷脂和磷脂酶的动态变化及其相互关系。高效薄层层析发现肝中DAG在第8周出现第一高峰,以后维持高于正常的水平,14周进一步增高,但以后下降。磷脂组分测定发现只有磷脂酰胆碱(PC)和肌醇磷脂类(PIs)在诱癌过程中降低,有可能成为DAG的来源,但PC可能是主要的,因PIs减少的量远小于DAG增加的量。进一步用酶偶联比色法测定PC专一性磷脂酶C(PC-PLC)和磷脂酶D(PLD)的活力,发现两酶在第8周的明显升高可能和DAG的第一高峰有关,而DAG在第18周的第二高峰可能主要来源于PC-PLC对PC的水解,PLD活力在10周后的逐步下降可能是14周后DAG的下降的原因之一,但尚有其他未知因素参与。对PC-PLC和PLD受PKC和癌基因的调节作了讨论。
The dynamic changes of 1,2-diglyceride (DAG), phospholipids, and phospholipase during the rat liver cancer induced by diethylnitrosamine were studied. High-performance thin-layer chromatography revealed that the first peak of DAG appeared in the liver during the 8th week, and maintained at a higher level than normal after 14 weeks, but decreased later. The determination of phospholipid components found that only phosphatidylcholine (PC) and inositol phospholipids (PIs) were reduced during the process of carcinogenesis, which may be the source of DAG, but PC may be the main reason, because the reduction of PIs is much smaller than that of DAG. Increase the amount. The activity of PC-specific phospholipase C (PC-PLC) and phospholipase D (PLD) was further determined by enzyme-coupled colorimetry. It was found that the apparent increase of the two enzymes in the 8th week may be related to the first peak of DAG. The second peak of DAG in the 18th week may come mainly from PC-PLC hydrolysis of PC. The gradual decline of PLD activity after 10 weeks may be one of the reasons for the decline of DAG after 14 weeks, but there are still other unknowns. Factors involved. The regulation of PC-PLC and PLD by PKC and oncogenes is discussed.
