采用荧光光谱法、微分脉冲伏安法和紫外可见光谱法相结合研究了那格列奈与牛血清白蛋白(BSA)的相互作用机制。考察了不同环境温度和酸度那格列奈与牛血清白蛋白的结合常数和结合位点,并对其热力学常数进行了研究。结果表明,两者结合距离为4.29 nm,那格列奈主要通过疏水作用和静电作用与牛血清白蛋白相作用,造成牛血清白蛋白二级结构的变化。静态猝灭和非辐射能量转移是导致牛血清白蛋白荧光猝灭的原因。此外,乙醇会对二者的结合作用产生影响。为那格列奈在生物体内的储存、传输及药理作用提供一些重要的信息。
The interaction between nateglinide and bovine serum albumin (BSA) was studied by fluorescence spectroscopy, differential pulse voltammetry and UV-Vis spectroscopy. The binding constants and binding sites of nateglinide and bovine serum albumin at different ambient temperatures and acidities were investigated and their thermodynamic constants were studied. The results showed that the binding distance between the two was 4.29 nm. Nateglinide interacted with bovine serum albumin mainly through hydrophobic interaction and electrostatic interaction, resulting in the change of secondary structure of bovine serum albumin. Static quenching and non-radiative energy transfer are the causes of fluorescence quenching of bovine serum albumin. In addition, ethanol will have an impact on the combination of the two. Provide some important information for the storage, transmission and pharmacological action of nateglinide in the organism.
