沉默信息调节因子1(sirtuin Type 1,SIRT1)是酵母Sir2的哺乳动物同源体,是一种NAD+依赖的Ⅲ类组蛋白去乙酰化酶,能使组蛋白和非组蛋白去乙酰化,参与细胞多种生理、病理过程,如基因沉默、细胞衰老、寿命延长、抗氧化应激、能量代谢调节、DNA损伤修复、肿瘤发生等。其中,SIRTl在乳腺癌的发生中涉及的上下游信号分子众多,信号通路繁杂而交叉。有研究显示SIRTl起着抑制乳腺癌发生的作用;而有的研究结果正相反,SIRTl起着促进乳腺癌发生的作用。
