目的:建立鞣花酸片血药浓度的高效液相色谱测定方法,并进行该药物在家兔体内的药代动力学研究。方法:采用Agilent C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈-0.1%磷酸水(23∶77)为流动相,流速0.8 mL.min-1,检测波长254nm,柱温30℃。结果:家兔血浆中,鞣花酸的线性范围为0.013~1.635 mg.L-1(r=0.998 9),方法回收率>95%,最低检测限为0.010 mg.L-1,家兔口服给药后,鞣花酸的药时曲线出现双峰,在家兔体内存在肝肠循环。结论:实验建立了家兔血浆中鞣花酸含量的HPLC测定方法,专属性强,可用于该药在家兔体内的药动学研究。
Objective: To establish a method for the determination of plasma concentration of ellagic acid tablets by high performance liquid chromatography (HPLC) and to study the pharmacokinetics of the drug in rabbits. Methods: The mobile phase was acetonitrile-0.1% phosphoric acid water (23:77) at a flow rate of 0.8 mL · min-1 on an Agilent C18 column (4.6 mm × 250 mm, 5 μm) with a detection wavelength of 254 nm and a column temperature of 30 ° C. Results: The linear range of ellagic acid in rabbits was 0.013-1.635 mg · L-1 (r = 0.998 9). The recovery rate was> 95% and the lowest detection limit was 0.010 mg.L-1 After administration, ellagic acid when the drug curve bimodal, in the rabbit there is enterohepatic circulation. Conclusion: The method for the determination of ellagic acid in rabbit plasma by HPLC was established experimentally. The method has high specificity and can be used to study the pharmacokinetics of the drug in rabbits.
