为探讨内源性 TGFα在创面的动态表达与分布,并分析其在创面愈合过程中的作用,采用mRNA 杂交技术与免疫组织化学技术,分别从基因与蛋白水平进行了研究。结果表明:创面愈合过程中存在内源性 TGFα的表达,且修复细胞迁移与增殖最活跃期间 TGFα的表达最强,当上皮化趋于完成时,其表达水平下降。EGF 受体的表达也呈与 TGFα相似的变化。免疫组化研究结果表明,内源性 TGFα主要分布于迁移、增殖的上皮细胞,其免疫染色的强弱与上皮细胞的生长、迁移呈正相关。提示:热损伤能诱导创面组织内源性 TGFα及其受体的表达,其表达存在受控机制并与修复细胞的迁移增殖有关。TGFα对上皮细胞发挥正性调节作用,主要参与表皮的修复。
In order to investigate the dynamic expression and distribution of endogenous TGFα in the wound and to analyze the role of endogenous TGFα in the process of wound healing, mRNA and protein levels were studied respectively by mRNA hybridization and immunohistochemistry. The results showed that there was endogenous TGFα expression during wound healing, and the expression of TGFα was the highest during the period of repairing cell migration and proliferation. When epithelialization tended to be completed, its expression level decreased. EGF receptor expression also showed a similar change with TGFα. Immunohistochemical results showed that endogenous TGFα mainly distributed in the migration and proliferation of epithelial cells, the intensity of immunostaining and epithelial cell growth and migration was positively correlated. It is suggested that heat injury can induce the expression of endogenous TGFα and its receptor in wound tissues. The expression of TGFα and its receptor exists in a controlled mechanism and is related to the migration and proliferation of repaired cells. TGFα play a positive regulatory role on epithelial cells, mainly involved in the repair of the epidermis.
