目的 探讨用高效液相(HPLC)PICO-TAG分析法测定正常成年人眼内玻璃体、晶状体、视网膜和色素膜内17种游离和水解氨基酸含量。方法 将以上组织标本依不同要求分别制成游离或水解组织标本,采用A,B双泵梯度法洗脱,以 Rom VERSION 3.86软件系统对各峰面积分析。结果 该方法对晶状体、视网膜和色素膜各组织内水解氨基酸的分离效果均很好,对玻璃体不论是水解还是游离样本,除对缬氨酸、蛋氨酸和胱氨酸无法将这3种氨基酸分离开,对其他氨基酸的分离效果均不错。结论 首次对我国正常成年人眼内组织17种游离和水解氨基酸做了测定,并提供了一种HPLC眼组织游离和水解氨基酸样本的制备方法。
Objective To investigate the content of 17 kinds of free and hydrolyzed amino acids in vitreous, lens, retina and pigmented membrane of normal adults by HPLC with PICO-TAG method. Methods The above tissue specimens were made into free or hydrolyzed tissue specimens according to different requirements, and were eluted by A and B double-pump gradient methods. The peak area was analyzed by Rom VERSION 3.86 software system. Results The method showed good separation effect on hydrolyzed amino acids in the tissues of the lens, retina and pigmented membrane. Whether the sample was hydrolyzed or free from vitreous, the three amino acids could not be separated from valine, methionine and cystine , The separation of other amino acids are good. Conclusion For the first time, 17 kinds of free and hydrolyzed amino acids in the intraocular tissues of normal adults in China were determined, and a method for preparing free and hydrolyzed amino acids in the ocular tissues of the eyes was provided.
