p34~(cdc2)是一种高度保守的34kD蛋白激酶亚单位,为G_2/M期转换的关键调节子。在有丝分裂细胞中,p34~(cdc2)与一种B型周期素结合,共同形成能被苏氨酸161磷酸化和苏氨酸去磷酸化激活的成熟促进子(MPF),活化的MPF诱导染色体聚集,破坏核膜,装配纺锤体。没有活化的p34~(cdc2),则细胞不能分裂。因此,对p34~(cdc2)在正常及肿瘤组织中的分布和表达水平的研究,有助于评价恶性肿瘤细胞周期调控情况。本研究用4种抗非洲蟾蜍属(Xenopus)p34~(cdc2)蛋白三分之二C-末端的单克隆抗体检查了p34~(cdc2)在人口腔正常、增生、恶变上皮及淋巴样组织的表达。
p34 ~ (cdc2) is a highly conserved 34 kD protein kinase subunit that is a key regulator of G 2 / M phase transition. In mitotic cells, p34 ~ (cdc2) binds to a type B cyclin and forms a mature promoter (MPF) that can be activated by threonine 161 phosphorylation and threonine dephosphorylation. The activated MPF induces chromosomes Aggregate, destroy the nuclear membrane, the assembly spindle. Without activated p34 ~ (cdc2), the cells can not divide. Therefore, the research on the distribution and expression of p34 ~ (cdc2) in normal and tumor tissue will help to evaluate the cell cycle regulation of malignant tumor cells. In this study, p34 ~ (cdc2) was examined in four human oral normal, hyperplastic, malignant epithelial and lymphoid tissues using four monoclonal antibodies against two-thirds of the C-terminus of Xenopus p34 ~ (cdc2) expression.
