本文采用了离心分离的方法,简单易行,可以同时分离到伯氏疟原虫的裂殖体、裂殖子和各感染期的红细胞,对于该原虫的实验研究有一定价值。本法首先从感染伯氏疟原虫的ICR系小鼠心脏采血,通过离心沉淀法洗净,由玻璃珠柱和纤维素粉柱除去白细胞和血小板,然后离心取得红细胞悬液。另将Isopaque(?)280和Munthe-Kaas&Seglen(1974)缓冲液、胎牛血清,配制成比重为1.320~1.082的10种梯度的溶液。再将红细胞悬液用10种梯度的溶液进行离心分离,红细胞悬液被分为8条带层,各带层中分别集中了感染疟原虫各期的红细胞和裂殖子等: 带层Ⅰ,比重1.200,未受染红细胞占86.3%;带层Ⅱ,比重1.158,环状体占80.2%;带层Ⅲ,比重1.147,大滋养体占57.8%、环状体占36.6%;带层Ⅳ~Ⅵ,比重分别为
In this paper, the method of centrifugation is simple and easy, and the schizonts, merozoites and erythrocytes of each stage of Plasmodium berghei can be isolated at the same time, which has some value for the experimental study of the protozoa. In this method, blood is collected from the heart of an ICR mouse infected with Plasmodium berghei and then washed by centrifugation. Leukocytes and platelets are removed by a glass bead column and a cellulose powder column, and then a suspension of erythrocytes is obtained by centrifugation.
