背景:丹参可有效减轻卡那霉素对CBA小鼠的耳毒性效应;然而,其是否也能防护庆大霉素对豚鼠的耳蜗毒性,目前尚不明确。目的:研究丹参注射液对庆大霉素耳蜗毒性的防护作用。设计:完全随机对照的实验研究。地点和对象:实验在中国医科大学听力研究室进行。选用清洁级健康纯白红目豚鼠40只,雌雄不限,体质量250~300g,耳郭反射灵敏,鼓膜及外耳道正常,由中国医科大学第二医院动物中心提供。干预:动物随机分成4组,即对照组、庆大霉素组、庆大霉素+丹参注射液组和丹参注射液组,每组各10只。庆大霉素组每日腹腔注射硫酸庆大霉素100mg/kg;丹参注射液组每日腹腔注射丹参注射液6g/kg;庆大霉素+丹参注射液组同时注射硫酸庆大霉素100mg/kg和丹参注射液6g/kg;对照组则每日腹腔注射等量生理盐水。4组皆连续用药10d。用药期间每天监测体质量以调整药量。全部实验由作者和另一名硕士研究生完成。主要观察指标:应用听脑干反应(auditorybrainstemresponse,ABR)测试,观察用药前后听阈变化。应用透射及扫描电镜技术,观察豚鼠耳蜗形态学变化。结果:用药10d后,庆大霉素组ABR阈值(36.00±5.48)dBSPL明显升高,与对照组(17.00±4.47)dBSPL比较差异有显著性意义(t=6.008,P<0.01);庆大霉素+丹参注射液组ABR阈值27.00±4.47)dBSPL较(庆大
BACKGROUND: Radix Salviae Miltiorrhizae can effectively reduce the ototoxic effect of kanamycin on CBA mice; however, it is still not clear whether it can also protect the cochlear toxicity of gentamicin against guinea pigs. Objective: To study the protective effect of Danshen injection on gentamicin cochlear toxicity. Design: A fully randomized controlled experimental study. Location and Subjects: Experiments were conducted at the Hearing Laboratory, China Medical University. Selection of clean grade healthy white red guinea pigs 40, male or female, body weight 250 ~ 300g, agar reflection sensitive, normal tympanic membrane and external auditory canal, provided by the Animal Center of the Second Hospital of China Medical University. Intervention: The animals were randomly divided into 4 groups: control group, gentamicin group, gentamicin + Danshen injection group and Danshen injection group, with 10 rats in each group. The gentamicin group was intraperitoneally injected with 100 mg / kg gentamicin sulfate per day; the Salvia miltiorrhiza injection group was intraperitoneal injected with 6 g / kg Salvia miltiorrhiza injection; the gentamicin plus salvia miltiorrhiza injection group was simultaneously injected with 100 mg gentamicin sulfate / kg and Salvia injection 6g / kg; control group were injected intraperitoneally with the same amount of saline. 4 groups were continuous medication 10d.
