目的建立沉香药材中色酮类成分含量测定方法,考察不同白木香结香方法的药材质量,为评价沉香质量提供实验依据。方法采用可见分光光度法(VS)以磷钨酸钼-硫酸锂为显色剂,以Aquilarone E色酮作为对照品溶液,在760 nm波长下测定沉香药材试液中沉香总色酮的含量。HPLC法测定沉香5种色酮含量,采用Waters Symmetry Shield C18色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈(A)-0.1%磷酸水(B),流速1m L·min-1,检测波长为252 nm,柱温30℃。结果所建立的可见分光光度法在5 h内稳定性良好,加样回收率为96.71%(RSD=2.12%)。HPLC法测定5种色酮含量在进样量0.1432~7.16、0.00884~0.442、0.01536~0.768、0.01264~0.632、0.0154~0.77μg范围内与峰面积积分值呈良好线性关系。5种色酮平均加样回收率分别为96.98%、97.19%、96.19%、96.52%及95.69%。不同批次沉香含量差异显著,与自然结香组(对照)比较,差异有统计学意义(P<0.05,P<0.01),人工诱导组白木香的结香质量优于传统火烫法,与结香周期较长的自然结香组相近。结论两种检测方法均简便、快速、准确,重复性好,可用于沉香中色酮类成分含量测定。人工诱导白木香快速结香可作为进一步开发利用其资源的重要方法。
OBJECTIVE To establish a method for the determination of the content of chromone in aloes medicinal materials, and to study the quality of medicinal materials of different almonds, and to provide the experimental basis for the evaluation of aloe quality. Methods The spectrophotometric method (VS) was used to determine the total flavonoids content of the radix pseudostellariae in test solution of Aquilaria sinensis by using molybdophosphotungstate-lithium sulfate as chromogenic agent and Aquilarone E chromone as reference solution. The HPLC method was used to determine the content of 5 kinds of chromones in Acacia confusa, using a Waters Symmetry Shield C18 column (4.6 mm × 250 mm, 5 μm) with a mobile phase of acetonitrile-A 0.1% phosphoric acid (B) and a flow rate of 1 mL · min -1 , Detection wavelength of 252 nm, column temperature 30 ℃. Results The visible spectrophotometry established within 5 h had a good stability. The average recovery was 96.71% (RSD = 2.12%). HPLC method for the determination of five kinds of chromone content in the injection volume 0.1432 ~ 7.16,0.00884 ~ 0.442,0.01536 ~ 0.768,0.01264 ~ 0.632,0.0154 ~ 0.77μg range and peak area integral value showed a good linear relationship. The average recoveries of five chromones were 96.98%, 97.19%, 96.19%, 96.52% and 95.69%, respectively. The contents of radix aloe in different batches were significantly different from those of the control group (P <0.05, P <0.01) Xiang Xiang longer natural incense group similar. Conclusion Both methods are simple, rapid, accurate and reproducible, and can be used to determine the content of chromone in aloe. Artificial induction of white wood incense quickly knot incense can be used as an important way to further develop their resources.
