目的观察不同剂量奥沙利铂对小鼠皮下移植肿瘤的抑制效果。方法建立胃癌小鼠模型,随机分3组:5%葡萄糖20 mL.kg-1对照组,小、大剂量(1.5,15.0 mg.kg-1)奥沙利铂组,腹腔等容积注射给药,连续10 d,第11 d处死小鼠,计算瘤重抑制率、肿瘤微血管密度,检测caspase-3活性,用Westernblot法检测VEGF和Ang-2的表达。结果小、大剂量(1.5,15.0 mg.kg-1)奥沙利铂组的抑瘤率分别为44.7%和15.8%(P<0.05)。2个剂量奥沙利铂组的caspase-3活性较对照组明显增加;但是小剂量奥沙利铂的caspase-3活性高于大剂量组(P<0.05)。小剂量奥沙利铂的的微血管密度、VEGF和Ang-2的表达较对照组明显下降(P<0.05)。结论小剂量奥沙利铂抑瘤作用明显,并具有抗肿瘤血管新生的作用。
Objective To observe the inhibitory effect of different doses of oxaliplatin on the subcutaneous tumor in mice. Methods The gastric cancer model was established and randomly divided into 3 groups: 5% glucose 20 mL.kg-1 control group, small and large dose (1.5, 15 mg.kg-1) oxaliplatin group, intraperitoneal injection The mice were sacrificed on the 11th day for 10 days. The tumor weight inhibition rate and tumor microvessel density were calculated. The activity of caspase-3 was detected. The expressions of VEGF and Ang-2 were detected by Western blot. Results The inhibitory rates of small and large dose (1.5,15.0 mg.kg-1) oxaliplatin group were 44.7% and 15.8% (P <0.05). The caspase-3 activity in the two doses of oxaliplatin group was significantly higher than that in the control group, but the activity of caspase-3 in the low-dose oxaliplatin group was higher than that of the high-dose oxaliplatin group (P <0.05). Small doses of oxaliplatin microvessel density, VEGF and Ang-2 expression was significantly decreased compared with the control group (P <0.05). Conclusion Low-dose oxaliplatin has obvious anti-tumor effect and anti-tumor angiogenesis effect.
