金属蛋白酶组织抑制因子 3(TIMP 3)是一种结合细胞外基质 (ECM)的非可溶性蛋白。TIMP 3参与正常组织的发育和重建过程 ,也参与了许多疾病的发生。TIMP 3一方面可以刺激成纤维细胞增殖 ,另一方面却诱导恶性肿瘤细胞的凋亡。由于TIMP 3能以 1∶1克分子比例与基质金属蛋白酶 (MMPs)非共价结合 ,抑制MMPs的活性 ,因此TIMP 3有抑制肿瘤生长、侵袭和转移的作用 ,肿瘤组织中TIMP 3表达水平的下调或丢失将促进肿瘤的进程。TIMP 3基因启动子区甲基化是其表达水平下调或丢失的主要原因。运用去甲基化因子可使TIMP 3启动子区去甲基化 ,使TIMP 3得以重新表达 ,这可能成为肿瘤治疗的新途径
Metalloproteinase Tissue Inhibitor 3 (TIMP 3) is a non-soluble protein that binds to the extracellular matrix (ECM). TIMP 3 is involved in the development and reconstruction of normal tissues and is involved in many diseases. TIMP 3 can stimulate fibroblast proliferation on the one hand, and induce apoptosis of malignant tumor cells on the other hand. Since TIMP 3 can non-covalently bind to matrix metalloproteinases (MMPs) in a 1: 1 molar ratio and inhibit the activity of MMPs, TIMP 3 has the effect of inhibiting tumor growth, invasion and metastasis. The expression of TIMP 3 in tumor tissues Downregulation or loss will promote tumor progression. TIMP 3 gene promoter methylation is the main reason for its down-regulation or loss of expression. Demethylation of the promoter region of TIMP3 using demethylation factors renders TIMP3 re-expressed, which may be a new approach to cancer therapy
