目的探讨2型糖尿病患者外周循环的内皮祖细胞(EPC)数量、功能与大血管病变的关系。方法 89例2型糖尿病患者按有无大血管病变分为大血管病变(MCV)组和非大血管病变(NMCV)组,27名健康人为正常对照(NC)组。经流式细胞分析循环EPC数,同时采血进行EPC的分离培养,7d后鉴定并检测增殖及迁移能力,对各组的一般临床资料,循环EPC数量、迁移、增殖能力指标及HOMA-IR等进行统计学分析。结果 MCV组循环EPC数量少于NMCV组。HOMA-IR及HbA_1c与循环EPC数量呈负相关。MCV组EPC的增殖能力和迁移能力均低于NMCV组。结论胰岛素抵抗和高血糖可能部分通过损害循环EPC的数量及功能,从而影响糖尿病大血管并发症的形成。
Objective To investigate the relationship between the number of circulating endothelial progenitor cells (EPCs) and the function of macrovascular vessels in type 2 diabetic patients. Methods Eighty - nine patients with type 2 diabetes mellitus were divided into two groups based on the presence or absence of macrovascular disease: macrovascular disease (MCV) and non - macrovascular disease (NMCV), and 27 healthy controls were included in the control group. The number of circulating EPCs was analyzed by flow cytometry. At the same time, the blood was collected for the isolation and culture of EPCs. After 7 days, the proliferation and migration ability were identified and tested. The general clinical data, number of circulating EPCs, migration and proliferation index and HOMA-IR Statistical analysis. Results The number of circulating EPC in MCV group was less than that in NMCV group. HOMA-IR and HbA_1c were negatively correlated with the number of circulating EPCs. The proliferation and migration ability of EPC in MCV group were lower than those in NMCV group. Conclusion Insulin resistance and hyperglycemia may partly affect the formation of diabetic macrovascular complications by impairing the number and function of circulating EPCs.
