目的研究大豆异黄酮及雌马酚的雌激素样作用在DMBA诱导的去势大鼠乳腺肿瘤发生中的作用。方法时间分辨荧光免疫分析法检测血清中雌二醇、雌三醇含量,Western-blot法和RT-PCR法检测乳腺组织中ERα、ERβ蛋白及基因表达。结果 SI、雌马酚各剂量组乳腺肿瘤发生率显著低于阳性对照组;SI高、中剂量(1000、500 mg/kg饲料)组和雌马酚高、中剂量组(400、200mg/kg饲料)雌二醇含量显著高于阳性对照组,雌三醇差别无统计学意义(P>0.05);SI和雌马酚各剂量组ERβ基因及蛋白表达显著高于阳性对照组(P<0.05);SI低剂量组ERα基因及蛋白表达显著低于阳性对照组(P<0.05)。结论 SI和雌马酚能够降低去势大鼠乳腺肿瘤发病率,其作用可能与改变内源性雌激素水平及乳腺组织ER表达有关。
Objective To investigate the role of estrogen-like action of soy isoflavones and equol in the development of mammary tumors induced by DMBA in ovariectomized rats. Methods The levels of estradiol and estriol in serum were detected by time-resolved fluorescence immunoassay. The expressions of ERα, ERβ and ERβ in breast tissues were detected by Western-blot and RT-PCR. Results The incidences of breast tumors in SI and equol groups were significantly lower than those in the positive control group. High and medium doses of SI and medium (1000, 500 mg / kg) and high and medium doses of equol (400 and 200 mg / kg) Dietary) estradiol content was significantly higher than the positive control group, estriol difference was not statistically significant (P> 0.05); SI and equol doses ERβ gene and protein expression was significantly higher than the positive control group (P <0.05 ); SI low-dose group ERα gene and protein expression was significantly lower than the positive control group (P <0.05). Conclusion SI and equol can reduce the incidence of breast tumors in ovariectomized rats, which may be related to the changes of endogenous estrogen levels and the expression of ER in breast tissues.
