目的和方法:应用免疫荧光及免疫电镜技术,对慢性酒精投与之实验大鼠及重症酒精性肝炎患者肝脏枯否细胞对LPS的清除功能进行了观察。结果:经门静脉注射LPS10min后,Lieber’s酒精饮食投与6周之大鼠肝脏枯否细胞内抗LPS荧光显色低于正常大鼠,电镜下枯否细胞内抗LPS阳性颗粒少见,而肝窦壁内皮细胞及肝细胞内抗LPS阳性物增多。重症酒精性肝炎患者肝穿组织枯否细胞内抗LPS荧光显色亦显著低于正常对照。结论:酒精刺激后肝脏枯否细胞对LPS的清除功能降低,且内毒素可能参与酒精致肝损伤的发生机制
PURPOSE AND METHODS: The clearance of LPS by hepatic Kupffer cells from experimental rats administered with chronic alcohol and patients with severe alcoholic hepatitis was observed by immunofluorescence and immunoelectron microscopy. Results: LPS fluorescence staining of Kupffer cells in the liver of Kupffer rats fed with Lieber’s alcohol diet for 6 weeks was lower than that of normal rats after intravenous injection of LPS for 10 minutes. The anti-LPS positive particles were rare in Kupffer cells under electron microscope, Wall endothelial cells and hepatocytes increased anti-LPS-positive. Severe alcoholic hepatitis patients with liver puncture Kupffer cells intracellular anti-LPS fluorescence color was also significantly lower than the normal control. CONCLUSION: Liver Kupffer cells can reduce the clearance function of LPS after alcohol stimulation, and endotoxin may be involved in the pathogenesis of alcohol-induced liver injury
