目的 鉴定含单纯疱疹病毒 (Herpessimplexvirus ,HSV)糖蛋白C(gC)模拟表位 (Mimotope)的短肽的免疫原性 ,确定其作为新型短肽疫苗组分的可能性。方法 将淘筛噬菌体随机肽库获得的递呈全部或部分 gC模拟肽的噬菌体分为 7组免疫动物 ,ELISA法检测噬菌体免疫效果及天然抗原免疫鼠阳性血清与噬菌体的反应性 ,观察诱生抗体持续时间。结果 递呈 gC模拟肽的噬菌体可诱导小鼠产生抗gC的抗体 ,抗体持续时间超过 4个月 ,用HSV天然抗原免疫小鼠获得的血清抗体也能与各组噬菌体结合。结论 噬菌体递呈肽能够模拟天然HSVgC抗原的部分免疫活性且具有一定的免疫原性 ,可用作HSV亚单位疫苗或表位疫苗的研究。
Objective To identify the immunogenicity of short peptides containing herpes simplex virus (HSV) glycoprotein C (gC) mimotope and to determine their potential as novel short peptide vaccine components. Methods The phages which were obtained by panning the phage random peptide library and presented all or part of the gC mimic peptide were divided into 7 groups. The phage immune effect and the reactivity of the phage antigen with the natural antigen immunized mice were detected by ELISA. duration. As a result, phage expressing gC peptidomimetic induced mouse anti-gC antibody for more than 4 months. Serum antibodies obtained from mice immunized with HSV native antigen also bound to phage. Conclusion The phage-presenting peptide can mimic the partial immunogenicity of the native HSV gC antigen and has certain immunogenicity, which can be used as a vaccine for HSV subunit or epitope vaccine research.
