目的探讨罗勒多糖(BPS)对卵巢癌SKOV3细胞血管内皮生长因子(VEGF)受体表达的调控作用及对其侵袭能力的影响。方法酶联免疫吸附试验(ELISA)、实时定量PCR及流式细胞术分别检测BPS对SKOV3细胞分泌VEGF的影响及对VEGF受体(VEGFRs)在SKOV3细胞表达的调控作用;Transwell试验检测不同浓度BPS对SKOV3细胞侵袭能力的影响;通过特异性shRNA慢病毒表达载体感染的方法稳定敲减VEGFR2在SKOV3细胞的表达后,Transw ell试验检测其对BPS侵袭抑制能力的影响。结果 BPS并未影响SKOV3细胞VEGF分泌水平的表达,对VEGFR1、VEGFR3在SKOV3的表达也无显著影响,但可明显上调VEGFR2在SKOV3细胞的表达;Transwell试验显示BPS可抑制SKOV3细胞的体外侵袭能力,并具有一定的浓度依赖性;成功感染携带VEGFR2 shRNA的慢病毒表达载体后,VEGFR2在SKOV3细胞的表达显著下调(P<0.01),且BPS对敲减VEGFR2表达的SKOV3细胞侵袭抑制能力显著降低。结论 BPS可显著抑制SKOV3细胞的侵袭能力,其抑制作用部分是通过上调VEGFR2在SKOV3细胞的表达介导的。
Objective To investigate the regulatory effect of basil polysaccharide (BPS) on the expression of vascular endothelial growth factor (VEGF) receptor in ovarian cancer SKOV3 cells and its effect on invasiveness. Methods The effect of BPS on the secretion of VEGF by SKOV3 cells and the regulation of the expression of VEGFRs in SKOV3 cells were detected by enzyme-linked immunosorbent assay (ELISA), real-time quantitative PCR and flow cytometry. Transwell assay was used to detect the expression of BPS On invasion ability of SKOV3 cells. Transfection of VEGFR2 in SKOV3 cells by shRNA shRNA lentivirus infection method was used to detect the effect of invasion inhibition on BPS. Results BPS did not affect the expression of VEGF in SKOV3 cells, but also had no significant effect on the expression of VEGFR1 and VEGFR3 in SKOV3 cells, but significantly up-regulated the expression of VEGFR2 in SKOV3 cells. Transwell assay showed that BPS inhibited SKOV3 cells in vitro invasion ability, The expression of VEGFR2 in SKOV3 cells was significantly down-regulated (P <0.01), and the inhibitory effect of BPS on SKOV3 cells knocked down VEGFR2 expression was significantly reduced after infection with lentiviral vector carrying VEGFR2 shRNA. Conclusion BPS can significantly inhibit the invasion of SKOV3 cells, and its inhibitory effect is partly mediated by up-regulating the expression of VEGFR2 in SKOV3 cells.
