目的:研究加味楂曲饮(JWZQY)干预非酒精性脂肪肝(NAFLD)的部分作用机制。方法:高脂饲料造模,空白组、模型组大鼠灌服等体积生理盐水,低、中剂量组分别灌服等体积含1 g/mL、2 g/mL浓度JWZQY溶液,荧光定量PCR法检测肝组织细胞色素C氧化酶Ⅰ(Cytochrome CoxidaseⅠ,COXⅠ)mRNA水平,Western Bloting法检测肝组织COXⅠ蛋白表达水平。结果:中剂量JWZQY干预后,大鼠COXⅠmRNA及蛋白水平均显著升高(P<0.01),与低剂量组比较,中剂量COXⅠmRNA及蛋白水平差异有统计学意义(P<0.01,P<0.05)。结论:JWZQY可增强COXⅠmRNA及蛋白表达,推测JWZQY防治非酒精性脂肪肝可能与影响线粒体功能有关。
Objective: To study the partial mechanism of JWZQY intervention on non-alcoholic fatty liver disease (NAFLD). Methods: The rats in the blank group and the model group were fed with the same volume of normal saline. The rats in the low dose and middle dose groups were dosed with the same volume of JWZQY solution at a concentration of 1 g / mL and 2 g / mL, respectively. The fluorescence quantitative PCR The level of cytochrome C oxidase Ⅰ (COX Ⅰ) mRNA in liver tissue was detected. The expression of COX Ⅰ protein in liver tissue was detected by Western Bloting. Results: Compared with the low dose group, the COX Ⅰ mRNA and protein levels in the middle dose of JWZQY were significantly increased (P <0.01, P <0.05) . Conclusion: JWZQY can enhance the expression of COX Ⅰ mRNA and protein, presumably JWZQY prevention and treatment of non-alcoholic fatty liver may be related to mitochondrial function.
