目的观察缰核损毁对下丘脑内GABAA受体α1亚单位(GABAAα1)表达的影响,以揭示缰核与下丘脑之间功能联系的可能的分子机制。方法建立缰核损毁大鼠模型,以假手术组为对照。分离下丘脑,提取总RNA及总蛋白,采用RT-PCR和Western blot方法检测两组大鼠下丘脑内GABAAα1 mRNA的转录水平及蛋白的表达水平。结果缰核损毁大鼠下丘脑内GABAAα1 mRNA的转录水平及蛋白表达水平均明显高于假手术对照组。结论缰核损毁可显著提高下丘脑内GABAAα1的表达水平,提示GABAA受体可能在缰核与下丘脑共同参与的生理调节过程中起重要作用。
Objective To investigate the effect of habenular nucleus lesion on the expression of GABAAα1 subunit in hypothalamus in order to reveal the possible molecular mechanism of the functional connection between habenula and hypothalamus. Methods Establishment of rat model of habenular lesion, sham operation group as control. The hypothalamus was isolated and the total RNA and total protein were extracted. The transcription level and protein expression of GABAAα1 mRNA in the hypothalamus were detected by RT-PCR and Western blot. Results The transcription level and protein expression of GABAAα1 mRNA in the hypothalamus of rats with habenular lesion were significantly higher than those of the sham operation control group. Conclusions The destruction of habenular nuclei can significantly increase the expression of GABAAα1 in the hypothalamus, suggesting that GABAA receptor may play an important role in the physiological regulation of habenular hypothalamic involvement.
