目的:病理学上诊断同样为肺鳞癌的患者,其治疗疗效和愈后往往存在一定差异,本研究利用本实验室制作的208个肺癌相关基因芯片,探讨了7例肺鳞癌组织中基因表达的异质性,试图为患者的治疗和预后提供分子依据。方法:提取7例男性肺鳞癌患者(年龄在55~65岁之间)癌组织的总RNA并用LD-PCR标记成探针,然后与含有208个肺癌相关基因的cDNAMicroarray杂交,芯片用ImaGene软件分析和处理数据。结果:7例肺鳞癌组织之间208个肺癌相关基因表达的相似性在60.65%~82.69%之间。结论:本研究首次对肺鳞癌患者不同个体之间基因的表达进行研究并发现存在一定的差异,提示应注意癌症患者的个体化诊断和治疗。
OBJECTIVE: Pathological diagnosis of patients with squamous cell carcinoma of the same lung often shows some differences in therapeutic efficacy and prognosis. In this study, 208 lung cancer related gene microarrays produced in our laboratory were used to investigate the relationship between the expression of genes The heterogeneity of expression, trying to provide the molecular basis for the treatment and prognosis of patients. Methods: Total RNA was extracted from 7 male patients with lung squamous cell carcinoma (aged 55-65 years old) and labeled with LD-PCR and then hybridized with cDNA Microarray containing 208 lung cancer related genes. The chip was analyzed with ImaGene software Analyze and process the data. Results: The expression of 208 lung cancer related genes among 7 cases of lung squamous cell carcinoma was 60.65% ~ 82.69%. Conclusion: This study for the first time in lung squamous cell carcinoma between different individuals to study the gene expression and found that there are some differences, suggesting that individualized diagnosis and treatment of cancer patients should pay attention.
