目的观察孕鼠羊水栓塞模型实验前后血清补体C3、C4和B因子的动态变化,探究羊水栓塞引发的类过敏反应。方法取健康妊娠晚期SD大鼠40只,随机分为两组,每组20只。两组孕鼠麻醉后从左侧股静脉取血,再暴露子宫抽取羊水。按2.5ml/kg由右侧股静脉缓慢注入自体羊水作为实验组,注入等剂量生理盐水作为对照组。注射1h后从右心房取血,将所取血样离心后分离血清,用ELISA法检测血清中补体C3、C4和B因子含量。结果羊水注射1h后血清补体C3、C4和B因子降低,差异有统计学意义(P<0.05)。结论孕鼠羊水栓塞补体系统被过度激活,消耗了补体成分,引发类过敏反应。
Objective To observe the dynamic changes of serum complement C3, C4 and B factors before and after the amniotic fluid embolism model in pregnant rats and explore the anaphylactoid reaction induced by amniotic fluid embolism. Methods Forty healthy SD pregnant rats were randomly divided into two groups (20 in each group). Two groups of pregnant rats after anesthesia blood from the left femoral vein, and then exposed to the uterus to extract amniotic fluid. According to 2.5ml / kg slow injection of autologous amniotic fluid from the right femoral vein as the experimental group, an equal dose of normal saline was injected as a control group. Blood samples were taken from the right atrium after injection for 1h, and the serum samples were separated by centrifugation. Serum complement C3, C4 and B were assayed by ELISA. Results After amniotic fluid injection for 1h, the levels of C3, C4 and B in serum complement decreased, with statistical significance (P <0.05). Conclusion The pregnant rat amniotic fluid embolism complement system is over-activated, consuming complement components and triggering anaphylactoid reactions.
