目的:探讨益智宁对注意缺陷多动障碍动物模型SHR大鼠学习记忆能力的影响。方法:将50只幼年SHR大鼠随机分为益智宁高、中、低剂量组、空白对照组、哌甲酯对照组,每组10只,益智宁高、中、低剂量组分别给予益智宁煎药液(14.4 g/kg,7.2 g/kg,3.6 g/kg),哌甲酯对照组给予哌甲酯(0.0015 g/kg),空白对照组给予生理盐水,每组均按12.5 m L/kg等容量灌胃给药。给药4周后,通过Morris水迷宫检测SHR大鼠学习记忆能力,定位航行实验5 d,空间探索实验1 d。结果:在定位航行实验中,空白对照组的逃避潜伏期表现不稳定,各用药组潜伏期随着训练时间的增加,均有逐渐缩短趋势,到训练第5天,各组间比较,差异有统计学意义(P<0.05);益智宁高剂量组、哌甲酯对照组逃避潜伏期明显缩短,与空白对照组比较,差异有统计学意义(P<0.05),但益智宁高剂量组与哌甲酯对照组比较,差异无统计学意义(P>0.05);在空间探索实验中,益智宁高剂量组在目标象限游泳的路程最长,与空白对照组比较,差异有统计学意义(P<0.05),哌甲酯对照组及益智宁高剂量组在目标象限游泳的时间及穿台次数均高于空白对照组,差异有统计学意义(P<0.05),但哌甲酯对照组与益智宁高剂量组比较,差异无统计学意义(P>0.05)。结论:益智宁高剂量及哌甲酯均能改善SHR大鼠的学习能力和记忆能力。
Objective: To investigate the effects of Yizhining on learning and memory abilities in SHR rats with attention deficit hyperactivity disorder. Methods: Fifty young SHR rats were randomly divided into Yizhienning high, medium and low dose groups, blank control group and methylphenidate control group, with 10 rats in each group. Yizhining high, medium and low dose groups were given Yizhining decoction liquid (14.4 g / kg, 7.2 g / kg, 3.6 g / kg), methylphenidate control group were given methylphenidate (0.0015 g / kg), and the blank control group was given normal saline, 12.5 m L / kg volumetric administration. Four weeks after the administration, the learning and memory abilities of SHR rats were detected by Morris water maze. The navigation experiment was located for 5 days and the space exploration experiment for 1 day. Results: In the navigation experiment, the escape latency of the blank control group was not stable. The incubation period of each medication group decreased gradually with the increase of the training time. By the fifth day of training, the differences between the two groups were statistically significant (P <0.05). Compared with the blank control group, the Yizhining high dose group and the methylphenidate control group showed a significantly shorter escape latency (P <0.05) (P> 0.05). In the space exploration experiment, Yizhining high dose group had the longest distance swimming in the target quadrant, compared with the blank control group, the difference was statistically significant (P> 0.05) P <0.05). The time of swimming in the target quadrant and the number of perforating times in the methylphenidate control group and the Yizhininging high-dose group were all higher than those in the blank control group (P <0.05), but methylphenidate control There was no significant difference between the high dose group and Yizhining (P> 0.05). Conclusion: Yizhining high dosage and methylphenidate can improve the learning ability and memory ability of SHR rats.
