[目的]为研究非乙烯敏感型菊科植物的木葡聚糖内糖基转移酶/水解酶(xyloglucan endo-transglycosylase/hydrolase,XTH)基因的结构特征。[方法]采用Trizol方法从菊花花瓣中提取总RNA,利用RT-PCR技术和T/A载体克隆XTH基因的cDNA序列。[结果]测序结果表明,克隆的cDNA片段大小为911bp,编码293个氨基酸残基,具有XTH蛋白家族的7个活性位点,命名该基因序列为CmXTH(基因登录号为HM752243);其推导的氨基酸序列与其它19种植物的XTH具有较高的同源性,其中与非洲菊、蕃茄的亲缘关系最近,而与猕猴桃、草莓、胡杨等的亲缘关系较远。[结论]克隆片段确为XTH基因的cDNA序列,可能与菊花花瓣的生长与衰老过程有关。
[Objective] The research aimed to study the structural characteristics of xyloglucan endo-transglycosylase / hydrolase (XTH) gene in xyloglucan of non-ethylene sensitive composites. [Method] Trizol method was used to extract total RNA from chrysanthemum petals. The cDNA sequence of XTH gene was cloned by RT-PCR and T / A vector. [Result] The sequencing results showed that the cloned cDNA fragment was 911bp in size and encoded 293 amino acid residues, which contained 7 active sites of XTH protein family and named CmXTH (Gene Accession Number HM752243). The deduced The amino acid sequence has high homology with XTH of other 19 species of plants, of which Gerbera and tomato have the closest genetic relationship, while kiwifruit, strawberry and Populus euphratica are more distantly related. [Conclusion] The cloned fragment was indeed the cDNA sequence of XTH gene, which may be related to the growth and senescence process of the petal chrysanthemum.
