抑制复制蛋白A1表达对食管癌细胞放射敏感性的影响

目的 观察抑制复制蛋白A1(RPA1)表达对食管癌细胞放射敏感性的影响.方法 脂质体介导转染RPA1反义寡核苷酸及阴性对照寡核苷酸于人食管癌TE-1细胞中,设空白对照组(只加培养基)、Mock组(转染试剂组)、对照组(阴性对照寡核苷酸组)及实验组(反义寡核苷酸组),分别以24、48、72、96h为转染时间点;通过逆转录-聚合酶链反应(RT-PCR)、实时荧光定量PCR法(qRT-PCR)和Western blot法分别检测各组食管癌TE-1细胞RPA1 mRNA及其蛋白表达;并给予6MV X射线单次照射6Gy,用细胞计数试剂盒(CCK)-8法检测各组食管癌细胞放射敏感性的差异.结果 RT-PCR、qRT-PCR及Western blot法结果显示实验组食管癌TE-1细胞的RPA1 mRNA 及蛋白表达均降低;CCK-8结果显示实验组食管癌TE-1细胞的增殖率低于其他各组(P<0.05).结论 抑制食管癌rE-1细胞RPA1表达,可增加食管癌TE-1细胞的放射敏感性.
