本试验结果表明,A蛋白夹心酶联法检测苹果花叶病毒,适宜工作浓度第1抗血清为1/103-1/104,第2抗血清为1/103。双抗体夹心法检测该病毒,抗体IgG浓度愈高,阳性样品与阴性样品吸光值之比愈大,当IgG稀释400倍时,两者的吸光值无明显差别。取休眠期枝条进行检测,以含0.05%吐温-20的磷酸缓冲盐水(PBST)十2%聚乙烯吡咯烷酮十0.1%卵蛋白作提取缓冲液获得了较好的检测结果,添加铜试剂能明显提高阳性样品吸光值。春季嫩叶的检测效果最佳。
