目的:探讨人肺腺癌A549多重抗药细胞株生物学特性变化。方法:采用阿霉素(ADM)浓度递增的方法诱导人肺腺癌A549细胞系(A549/P),在体外持续培养6个月,获得了具有多重抗药性的A549/R2细胞,该细胞能在0.2μg/mlADM下持续增殖。通过光镜和电镜观察其增殖及形态结构变化。结果:A549/R2细胞由巨大细胞增殖而来,其微绒毛、线粒体及分泌泡明显增加,有进一步分化趋向。结论:A549/R2细胞的进一步分化特征可能与其抗药性形成有关。
Objective: To investigate the biological characteristics of human lung adenocarcinoma A549 multidrug resistant cell line. METHODS: Human lung adenocarcinoma cell line A549 (A549/P) was induced by increasing adriamycin (ADM) concentration. After continuous culture in vitro for 6 months, A549/R2 cells with multiple drug resistance were obtained. Proliferation was continued at 0.2 μg/ml ADM. The proliferation and morphological changes were observed by light and electron microscopy. RESULTS: A549/R2 cells were derived from giant cell proliferation, and their microvilli, mitochondria, and secretory vesicles were significantly increased and further differentiated. Conclusion: The further differentiation of A549/R2 cells may be related to the formation of drug resistance.
